I am interested in unravelling the evolutionary strategies employed in the formation of functional proteins from their gene sequence, and in ultimately exploiting this knowledge in a predictive manner in the development of new medicines.

 

Bioinformatics Tools for Membrane Proteins

Development of novel bioinformatics approaches, tools and resources for generic application to membrane protein sequences:

·         Data mining approaches for prediction of the subcellular location and function of membrane proteins from sequence (TMLocate and TMFun, with Gorka Lasso, CIC bioGune, Bilbao and John Antoniw, Rothamsted Research)

·         Knowledge based prediction of associations and relative positioning of transmembrane regions in multi-transmembrane region proteins (TMRelate, with Roberto Togawa, Embrapa, Brazil)

·         Sequence pattern discovery approaches for functional domains such as re-entrant loops, as found in the aquaglyceroporin family, potassium channels etc. (TMLoop, with Gorka Lasso and John Antoniw).

Protein Modelling

High throughput structural modelling of protein families and systematic in silico prediction of effects of gene mutations on protein structure and function:

·         Brain transport proteins and ion channels (with Prof. Mark Rees, Swansea)

·         Claudins and viral receptors (with Prof. Jane McKeating, Birmingham)

·         Cytochrome P450s (with Prof. Steve Kelly, Swansea)

 

The projects involve structural modelling of dozens of proteins and the effects of hundreds of mutations. My central question surrounds identification of common patterns in the way evolutionary pressures, expressed in gene coding, lead to specific changes in protein structure that result in normal and aberrant function.

 

Genome Bioinformatics

Structural and functional annotation of:

·         The membrane proteins of newly sequenced genomes, such as Fusarium graminearum, Mycosphaerella graminicola (with John Antoniw and Kim Hammond-Kosack, Rothamsted Research)

·          Cytochrome P450 proteins (CYPs) of Aspergillus nidulans (with Diane Kelly and Steve Kelly, Swansea)

 

Medicine Teaching

I greatly enjoy my lecturing on the Swansea Graduate Entry Medicine Programme. I lead the Nutrition Theme and I lecture on metabolism and clinical biochemistry, intestinal absorption and genetics.
